E6201 + dabrafenib is a combination of two small molecule kinase inhibitors under investigation for the treatment of BRAF V600-mutated metastatic melanoma, particularly with central nervous system (CNS) metastases. E6201 is an ATP-competitive Mitogen-activated protein kinase kinase 1 (MEK1) inhibitor that demonstrates potent activity in BRAF V600-mutant melanoma cell lines and has excellent brain distribution characteristics, making it promising for CNS tumors. Dabrafenib is a selective ATP-competitive inhibitor of mutant Serine/threonine-protein kinase B-Raf (BRAF) kinases (especially BRAF V600E), approved for use in several cancers with this mutation. The combination aims to target the MAPK pathway at multiple points to inhibit tumor growth and overcome resistance mechanisms[2][4][5][6][7][8].
