Drug intelligence / Profile preview
EA01 and EA02 are two novel tricyclic thioridazine derivatives developed by researchers at the University of Bergen and collaborating institutions. Designed to overcome the cardiotoxicity associated with the parent antipsychotic drug thioridazine, these compounds are being investigated for their antiproliferative, apoptosis-inducing, and chemosensitizing activities in glioblastoma multiforme (GBM). In preclinical in vitro studies, EA01 and EA02 demonstrated selective cytotoxicity against GBM cell lines and glioma stem cells (GSCs) at concentrations of 4 µM and 9.5 µM, respectively, while sparing normal human astrocytes and rat brain organoids. In vivo animal studies have shown that intravenous administration of these compounds up to 21 mg/kg is well-tolerated with limited systemic toxicity.
