ECI-006 + anti-PD-1 immunotherapy is an investigational combination regimen designed for the treatment of melanoma. ECI-006 is an mRNA-based cancer vaccine consisting of a combination of "TriMix" (mRNAs encoding dendritic cell activators: CD40L, CD70, and constitutively active TLR4) and mRNAs for multiple melanoma-associated tumor antigens (tyrosinase, gp100, MAGE-A3, MAGE-C2, PRAME). ECI-006 is administered intranodally to stimulate patient dendritic cells to initiate robust T-cell responses against melanoma antigens. The combination with anti-PD-1 immunotherapy (such as pembrolizumab or nivolumab) is intended to enhance the anti-tumor immune response by both priming tumor-specific T cells (via ECI-006) and preventing T-cell exhaustion (via PD-1 pathway blockade)[3][6]. Early-phase trials demonstrated that ECI-006 is immunogenic in a proportion of patients and generally well tolerated, and further development in combination with anti-PD-1 therapy is ongoing in melanoma[3][6].
