**ECI830 + ribociclib + fulvestrant** is a combination regimen under clinical investigation for the treatment of advanced hormone receptor-positive, human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer. ECI830 is a novel CDK2 inhibitor; ribociclib is a CDK4/6 inhibitor, and fulvestrant is an estrogen receptor antagonist. The combination targets the cell cycle and estrogen signaling: ECI830 inhibits cyclin-dependent kinase 2 (CDK2), ribociclib inhibits CDK4/6, and fulvestrant causes degradation and antagonism of estrogen receptor, collectively aiming to suppress tumor cell proliferation and overcome resistance to endocrine therapy. The regimen is being evaluated in phase I/II studies to assess safety, optimal dosing, and anti-tumor efficacy after progression on prior hormonal therapy and CDK4/6 inhibitor[1][3][4][7][10].
