EcNΔlpp::A5 is an engineered live probiotic based on **Escherichia coli Nissle 1917 (EcN)**. It is genetically modified to display **Annexin A5 (ANXA5)** on its surface, enhancing its targeting and colonization at sites of intestinal inflammation where phosphatidylserine (PS) is exposed on apoptotic colon epithelial cells. Additionally, the lipoprotein gene (**lpp**) is knocked out to increase secretion capability. EcNΔlpp::A5 acts by improved localization to inflamed mucosa and modulation of mucosal immune responses, showing efficacy in alleviating ulcerative colitis in preclinical models[2]. The strain forms the basis for subsequent therapeutic constructs (e.g., EcNΔlpp::A5-aTN, which further secretes anti-TNF-α nanobodies), but EcNΔlpp::A5 itself does not secrete anti-TNF agents[2]. Developed for **targeted drug delivery** and **microbiome therapy** primarily for **ulcerative colitis** and other inflammatory bowel diseases.
