**EcN-Apt@HG** is a research-stage, orally administered inflammation-targeted probiotic delivery system for ulcerative colitis. It comprises *Escherichia coli* Nissle 1917 modified on its surface with an interleukin-6-binding aptamer and encapsulated in a dynamic hydrogel formed from aldehyde-functionalized chondroitin sulfate and amine-terminated poly(amidoamine). The IL-6 aptamer promotes retention at inflamed intestinal sites, while hydrogel encapsulation protects bacterial viability during gastrointestinal transit and supports colonic colonization. In dextran sulfate sodium-induced colitis mouse models, the system restored mucosal barrier function, reduced hyperactive immune responses, remodeled gut dysbiosis, and improved depression-like behavior and cognitive impairment through gut-brain-axis-associated effects.
