Overview

EDD-University of Wisconsin, Madison-obesity refers to a specific experimental protocol and set of pharmacological probes used in a Phase 1 clinical trial (NCT02586311) conducted by the University of Wisconsin, Madison. The study evaluates microvascular function and endothelium-dependent dilation (EDD) in young obese adults compared to lean controls. The protocol utilizes a combination of vasodilatory agents—including acetylcholine, isoproterenol, bradykinin, and adenosine triphosphate (ATP)—to stimulate blood flow. To further investigate the underlying mechanisms of vasodilation, the study employs inhibitors such as NG-monomethyl-L-arginine (L-NMMA) to block nitric oxide synthase, ketorolac to block cyclooxygenase, and barium chloride to block inward-rectifier potassium channels. This comprehensive approach allows researchers to quantify the relative contributions of nitric oxide, prostaglandins, and endothelium-derived hyperpolarizing factors (EDHF) to vascular health in the context of obesity.
