EEDVsMit is an investigational nanocell-based targeted therapy developed by EnGeneIC Ltd. It utilizes the EnGeneIC Delivery Vehicle (EDV) platform, which consists of bacterially-derived nanocells approximately 400 nm in diameter. These nanocells are coated with bispecific antibodies (BsAb) where one arm recognizes the Epidermal Growth Factor Receptor (EGFR) on the tumor cell surface and the other arm binds to the EDV. The nanocells are loaded with the cytotoxic payload mitoxantrone, a type II topoisomerase inhibitor and DNA intercalator. Upon intravenous administration, the targeted EDVs bind to EGFR-expressing tumor cells, are internalized via endocytosis, and release the mitoxantrone directly into the intracellular environment. This delivery mechanism is designed to concentrate the chemotherapeutic agent within the tumor while reducing systemic exposure and associated toxicities. Clinical development, led by investigators such as Dr. David Ziegler, has focused on pediatric patients with recurrent or refractory solid tumors, central nervous system (CNS) tumors, and diffuse intrinsic pontine glioma (DIPG).
