**Eganelisib + nivolumab** is a combination therapy comprising eganelisib, a highly selective small molecule inhibitor of phosphoinositide 3-kinase gamma (PI3Kγ), and nivolumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). Eganelisib reprograms tumor-associated macrophages and reduces myeloid cell recruitment to the tumor microenvironment, thereby relieving immune suppression and enhancing immune activation. Nivolumab acts as an immune checkpoint inhibitor by blocking PD-1, restoring T-cell antitumor activity. The combination has been investigated primarily for metastatic urothelial carcinoma, head and neck squamous cell carcinoma, and melanoma, where it has shown the potential to improve response rates and disease control, especially in anti–PD-1/PD-L1-refractory or PD-L1–negative patient populations[1][3][5][7].
