Overview

A combination therapy consisting of an epidermal growth factor receptor (EGFR) monoclonal antibody and standard cytotoxic chemotherapy. EGFR antibodies such as cetuximab, panitumumab, or necitumumab are used to target the EGFR protein on cancer cells, inhibiting cell proliferation and survival signals. When combined with platinum-based or triplet chemotherapies (e.g., FOLFOXIRI), this regimen has demonstrated improved overall survival, progression-free survival, and objective response rates in several cancers—most notably metastatic colorectal cancer (mCRC) and non-small cell lung cancer (NSCLC). The anti-EGFR antibodies act by blocking ligand binding to the EGFR extracellular domain, preventing downstream signaling pathways involved in tumor growth. This combination is typically used as a first-line treatment for advanced or unresectable disease.
