EGFR BATs + temozolomide is an investigational combination therapy for the treatment of newly diagnosed grade 4 astrocytomas, including glioblastoma. The regimen consists of EGFR BATs (Bispecific Antibody Armed T cells) and the chemotherapy drug temozolomide. EGFR BATs are autologous, activated T cells that have been expanded ex vivo and coated with a bispecific antibody targeting both CD3 on the T cells and the epidermal growth factor receptor (EGFR) on tumor cells. This redirects the patient's own immune system to recognize and destroy EGFR-expressing glioma cells. Temozolomide is an oral alkylating agent that crosses the blood-brain barrier and induces DNA damage, leading to apoptosis in tumor cells. The combination is typically administered alongside standard-of-care radiation therapy to maximize anti-tumor efficacy and stimulate a systemic immune response against the malignancy.
