Overview

A combination therapy consisting of an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) and cytotoxic chemotherapy. EGFR-TKIs are small molecule inhibitors that block the intracellular tyrosine kinase domain of the EGFR, thereby inhibiting downstream signaling pathways involved in cell proliferation and survival. Chemotherapy agents used in this combination are typically platinum-based compounds (such as cisplatin or carboplatin), pemetrexed, gemcitabine, or paclitaxel, which act by interfering with DNA replication or mitosis to induce cancer cell death. This combination has been investigated primarily for non-small cell lung cancer (NSCLC), especially in patients with activating EGFR mutations. The rationale is to enhance anti-tumor efficacy and potentially overcome resistance mechanisms; however, clinical trials have shown mixed results regarding efficacy compared to monotherapy with either agent[1][2][3]. The approach may be used concurrently or sequentially as neoadjuvant, first-line, or maintenance therapy.
