ELENAGEN + gemcitabine is a combination therapy for **platinum-resistant ovarian cancer** that pairs ELENAGEN, a novel **DNA plasmid vaccine encoding the p62/SQSTM1 protein**, with gemcitabine, a standard chemotherapeutic agent. ELENAGEN operates through multiple mechanisms: it targets p62 (SQSTM1), a protein upregulated in various tumors and associated with poor prognosis and platinum resistance, by inducing antitumor T- and B-cell immune responses. Additionally, ELENAGEN alleviates chronic tumor-associated inflammation by suppressing proinflammatory cytokine generation (such as TNF, IL-1, IL-6), thereby enhancing chemotherapy efficacy. Its unique supercoiled circular DNA plasmid formulation facilitates immune cell infiltration into tumors (especially "cold tumors"), modifies the tumor microenvironment to promote tumor normalization, and sensitizes tumors to chemotherapy. No severe adverse events have been reported in clinical studies of ELENAGEN, whether as monotherapy or combined with gemcitabine[1][2][3].
