Overview

Elotuzumab + bortezomib is a combination therapy comprising two agents used in the treatment of relapsed or refractory multiple myeloma. **Elotuzumab** is a humanized IgG1 monoclonal antibody that targets signaling lymphocytic activation molecule F7 (SLAMF7), expressed on myeloma and natural killer cells. Its mechanism includes direct activation of natural killer (NK) cells and antibody-dependent cellular cytotoxicity (ADCC), as well as promoting antibody-dependent cellular phagocytosis (ADCP) by macrophages. **Bortezomib** is a small molecule proteasome inhibitor that disrupts protein degradation pathways in plasma cells, leading to apoptosis. Clinical studies have shown that combining elotuzumab with bortezomib (often also with dexamethasone) improves progression-free survival and deepens response rates without significant added toxicity compared to bortezomib-based regimens alone[1][2][4].
