Elotuzumab + lenalidomide + dexamethasone is a three-drug combination regimen used for the treatment of multiple myeloma, particularly in patients who have received one to three prior therapies. Elotuzumab is a humanized IgG1 monoclonal antibody that targets the Signaling lymphocyte activation molecule F7 (SLAMF7) protein on myeloma and natural killer (NK) cells, enhancing immune-mediated killing of myeloma cells. Lenalidomide is an immunomodulatory small molecule that exerts anti-myeloma effects through direct tumoricidal activity and immune modulation. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and cytotoxic properties against lymphoid malignancies. The combination has demonstrated improved progression-free survival and overall response rates compared to lenalidomide plus dexamethasone alone in relapsed or refractory multiple myeloma[1][2][6][8].
