Elranatamab + daratumumab is an investigational combination therapy for multiple myeloma, currently being studied primarily in relapsed/refractory and newly diagnosed transplant-ineligible patients. - **Elranatamab** is a humanized bispecific monoclonal antibody that targets B-cell maturation antigen (BCMA) on myeloma cells and CD3 on T-cells, thereby redirecting and activating T-cells to kill BCMA-expressing cancer cells. - **Daratumumab** is a human monoclonal antibody targeting CD38, a cell surface protein highly expressed on myeloma cells, leading to direct cell death and immune-mediated cytotoxicity through complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and antibody-dependent cellular phagocytosis. This combination has shown promising clinical activity and manageable safety profiles in ongoing phase 3 MagnetisMM studies for relapsed/refractory and newly diagnosed multiple myeloma[1][2][3][5][6].
