Overview

A combination regimen of **eltrombopag**, **daunorubicin**, and **cytarabine** would consist of three agents with distinct and complementary mechanisms of action: - **Eltrombopag** is an orally active, small molecule thrombopoietin receptor agonist that stimulates the production and differentiation of megakaryocytes, thereby increasing platelet counts. It is primarily used to treat thrombocytopenia in chronic immune thrombocytopenia or patients with chronic hepatitis C. - **Daunorubicin** is an anthracycline-class chemotherapy that intercalates DNA, inhibits topoisomerase II, and generates free radicals, causing DNA damage and cytotoxicity in rapidly dividing cells. - **Cytarabine** is a nucleoside analog antimetabolite that inhibits DNA synthesis, mainly active during the S-phase of cell division. It is a major agent in the treatment of acute myeloid leukemia (AML). The combination of daunorubicin + cytarabine (often as a liposomal formulation, e.g., CPX-351/Vyxeos) is FDA approved and is standard for certain high-risk AML subtypes[1][2][3][10]. Eltrombopag is not standardly combined with daunorubicin + cytarabine in AML and is primarily used for thrombocytopenia, but there is academic and clinical interest in minimizing thrombocytopenia during intensive chemotherapy.
