Overview

A combination therapy consisting of two thrombopoietin receptor agonists (eltrombopag and romiplostim) and a corticosteroid (prednisone). This combination has been used successfully in cases of severe thrombocytopenia, particularly in Evans syndrome and immune thrombocytopenic purpura (ITP) that have been refractory to standard treatments. The medications work through complementary mechanisms: eltrombopag binds to the transmembrane domain of the thrombopoietin receptor, romiplostim binds directly to the thrombopoietin binding site, and prednisone reduces autoimmune destruction of platelets.
