Overview

Eltrombopag + vorinostat is a combination of two small molecule drugs, each with distinct mechanisms of action and approved indications. Eltrombopag is a thrombopoietin receptor agonist that stimulates the bone marrow to produce more platelets, primarily used for treating thrombocytopenia in conditions such as chronic idiopathic thrombocytopenic purpura (ITP), severe aplastic anemia, and thrombocytopenia associated with chronic hepatitis C[2][5][8]. Vorinostat is a histone deacetylase (HDAC) inhibitor used mainly for cutaneous T-cell lymphoma (CTCL); it works by inhibiting class I and II HDAC enzymes, leading to increased acetylation of histones and other proteins, which can result in cell cycle arrest, apoptosis, inhibition of angiogenesis, and downregulation of immunosuppressive interleukins[3][4][6]. The combination may be explored in research or clinical settings where both platelet stimulation and epigenetic modulation are desired.
