ELVN-002 + trastuzumab is an investigational combination therapy composed of **ELVN-002**, a potent, highly selective, irreversible HER2 (human epidermal growth factor receptor 2) inhibitor, and **trastuzumab**, a monoclonal antibody targeting HER2. ELVN-002 is designed to inhibit both wild-type HER2 and key HER2 mutations, sparing wild-type EGFR to reduce EGFR-related toxicities. The combination is being studied to enhance internalization and ubiquitination of HER2, thereby promoting lysosomal degradation and increased efficacy especially when paired with antibody-drug conjugates (ADC) or other HER2-targeting therapies. Preclinical studies demonstrate that combining ELVN-002 with trastuzumab leads to greater HER2 receptor degradation and may overcome resistance in HER2+ solid tumors, including breast, colorectal, lung, and gastric cancers. ELVN-002 is being developed by Enliven Therapeutics and evaluated in Phase 1 trials in HER2+ solid tumors, with and without additional chemotherapeutic agents[1][2][3].
