**ELVN-002 + trastuzumab emtansine** is an investigational combination therapy for HER2-positive cancers, notably including HER2-positive metastatic breast cancer. The combination includes **ELVN-002**, a highly selective, irreversible small molecule tyrosine kinase inhibitor targeting the human epidermal growth factor receptor 2 (HER2), and **trastuzumab emtansine (Kadcyla)**, an antibody-drug conjugate that combines the anti-HER2 monoclonal antibody trastuzumab with the cytotoxic agent DM1 (emtansine). ELVN-002 enhances HER2 receptor internalization and degradation, potentially increasing the efficacy of HER2-targeted therapies. Trastuzumab emtansine targets HER2-expressing cancer cells, delivering DM1 directly to these tumor cells, which disrupts microtubule function and leads to cell death. The combination is being studied to improve anti-tumor effects through dual blockade and enhanced intracellular payload delivery specifically in HER2+ solid tumors, including metastatic breast cancer and possibly in patients with HER2 mutant non-small cell lung cancer[1][3][5].
