Overview

A four-drug combination of **emivirine (a non-nucleoside reverse transcriptase inhibitor, NNRTI)**, **stavudine (a nucleoside reverse transcriptase inhibitor, NRTI)**, **didanosine (an NRTI)**, and **hydroxyurea (an antimetabolite sometimes used experimentally to enhance antiretroviral efficacy)**, previously evaluated for the treatment of HIV-1 infections. - **Emivirine** is a failed experimental agent that acts as a NNRTI, binding allosterically to HIV-1 reverse transcriptase and inhibiting viral RNA-directed DNA polymerase activity[1][5][7][8]. - **Stavudine** and **didanosine** are NRTIs, inhibiting HIV-1 reverse transcriptase by competing with natural nucleosides[2]. - **Hydroxyurea** is a ribonucleotide reductase inhibitor, sometimes combined with NRTIs to potentially improve efficacy or resistance profiles[2]. This combination was studied for synergistic anti-HIV-1 activity. It showed potent inhibition of HIV-1 replication in vitro and had Phase 3 clinical trials involving emivirine plus NRTIs for HIV, but ultimately development was discontinued due to insufficient efficacy in humans[1][2].
