Empasiprubart (ARGX-117) is a first-in-class humanized monoclonal antibody that targets complement component C2. By binding to the Sushi-2 domain of C2, it blocks activation of both the classical and lectin pathways of the complement system while leaving the alternative pathway intact. This selective inhibition aims to reduce tissue inflammation and prevent complement-mediated cytotoxicity in autoimmune diseases without compromising antimicrobial defense provided by the alternative pathway. Recombinant human hyaluronidase PH20 (rHuPH20) is an enzyme used as a drug delivery technology to facilitate subcutaneous administration by enhancing dispersion and absorption of co-administered biologics such as monoclonal antibodies. The combination is being developed for autoimmune indications including multifocal motor neuropathy (MMN), delayed graft function after kidney transplantation, and dermatomyositis[1][4][5][7].
