Overview

This is a combination regimen of four oral drugs with distinct mechanisms of action used in oncology. Enasidenib is an inhibitor of mutant isocitrate dehydrogenase 2 (IDH2), primarily indicated for relapsed or refractory acute myeloid leukemia with IDH2 mutations. Dexamethasone is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressant properties. Ixazomib is an oral proteasome inhibitor that disrupts protein degradation pathways in cancer cells and is approved for use in multiple myeloma. Pomalidomide is an immunomodulatory agent that affects the tumor microenvironment and has direct antineoplastic effects; it also has approval for relapsed/refractory multiple myeloma. This specific four-drug combination does not have established clinical trial data as a single regimen; however, combinations including ixazomib, pomalidomide, and dexamethasone are under investigation for multiple myeloma[1][3][6]. The addition of enasidenib would be experimental.
