ENB-003 + pembrolizumab is a combination therapy under clinical investigation for advanced, refractory solid tumors, including platinum-refractory or resistant ovarian cancer, melanoma, and pancreatic cancer. ENB-003 is a first-in-class, selective small molecule antagonist of the endothelin receptor B (ETBR), a novel immune checkpoint implicated in immune evasion within the tumor microenvironment. Pembrolizumab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), widely approved as an immune checkpoint inhibitor. This combination is designed to overcome immunotherapy resistance by simultaneously inhibiting both ETBR-mediated and PD-1/PD-L1-mediated immune suppression, thus enhancing anti-tumor immune responses. The regimen is being evaluated in phase 1/2 clinical trials, and early results indicate promising response rates and disease control, especially in patient populations with limited options.
