Overview

**Encapsulated genetically engineered human cells + ifosfamide** is an investigational targeted chemotherapy platform developed by PharmaCyte Biotech for pancreatic cancer and potentially other solid tumors. Genetically modified human cells, engineered to express an enzyme (cytochrome P450), are encapsulated in **Cell-in-a-Box** polymer capsules and surgically implanted near the tumor in the blood supply. Administered intravenously at one-third the standard dose, the prodrug **ifosfamide** diffuses into the capsules where the cells convert it locally to its active cytotoxic form, **phosphoramide mustard**, enabling site-specific DNA alkylation and tumor cell death while minimizing systemic toxicity. Early clinical trials demonstrated safety, tolerability, tumor response (including conversion from inoperable to operable), and improved survival (e.g., median ~11 months vs. ~5.7 months with gemcitabine).[1][3]
