Overview

This is a combination regimen including **encorafenib**, a BRAF kinase inhibitor; **binimetinib**, a MEK kinase inhibitor; and **nivolumab**, an immune checkpoint inhibitor (PD-1 inhibitor). Encorafenib targets BRAF V600E/K mutant proteins in malignant cells, blocking cell proliferation driven by aberrant MAPK signaling. Binimetinib inhibits MEK1/2, further suppressing the MAPK/ERK signaling axis. Nivolumab is a human monoclonal antibody that binds PD-1 on T cells, preventing its interaction with PD-L1/PD-L2, thereby enhancing anti-tumor immune responses. This triple combination is under investigation for patients with advanced or metastatic melanoma with BRAF V600E or V600K mutations, aiming to leverage both targeted therapy against the MAPK pathway and immunotherapy to achieve deeper and more durable clinical responses[4][5][6][7].
