The combination of encorafenib, cetuximab, and FOLFIRI is an investigational regimen for the treatment of metastatic colorectal cancer (mCRC) with a BRAF V600E mutation. Encorafenib is a small molecule inhibitor targeting the BRAF V600E mutant kinase, while cetuximab is a monoclonal antibody that inhibits the epidermal growth factor receptor (EGFR). FOLFIRI is a chemotherapy regimen consisting of fluorouracil (a pyrimidine analog antimetabolite), leucovorin (folinic acid, which enhances fluorouracil activity), and irinotecan (a topoisomerase I inhibitor). This combination has shown promising antitumor activity and tolerability in patients with previously treated or untreated BRAF V600E-mutant mCRC in clinical trials[1][2][5]. The mechanism involves dual blockade of oncogenic signaling pathways along with cytotoxic chemotherapy to maximize tumor control.
