Overview

**endostatin + etoposide + carboplatin** is a combination regimen used in the treatment of cancers, notably small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC). **Endostatin** is an endogenous inhibitor of angiogenesis, working primarily by interfering with vascular endothelial growth factor (VEGF)-mediated pathways, thereby inhibiting tumor blood vessel formation and normalizing tumor vasculature. **Etoposide** is a topoisomerase II inhibitor that interferes with DNA synthesis, thus causing cell death in rapidly dividing tumor cells. **Carboplatin** is a platinum-based chemotherapeutic agent that alkylates and cross-links DNA, also inhibiting tumor cell replication. This regimen has been studied both in combination and sequentially, often with the aim to leverage endostatin’s anti-angiogenic activity to improve chemotherapeutic delivery and efficacy. Clinical studies (notably in China) suggest that adding endostatin to etoposide and carboplatin is tolerable, does not significantly worsen toxicity, and may offer improvement in quality of life and progression-free survival, although evidence for improvement in overall survival is limited[2][4].
