EndoTAG-1 + paclitaxel is a combination therapy consisting of EndoTAG-1 (paclitaxel embedded in positively-charged liposomal membranes) and standard paclitaxel. EndoTAG-1 specifically targets tumor endothelial cells that form blood vessels feeding cancer cells. The positively-charged lipids in EndoTAG-1 interact with negatively charged endothelial cells and release paclitaxel, effectively destroying the tumor's blood supply[3]. Paclitaxel itself is a mitotic inhibitor that works by interfering with normal cell division, making it an effective chemotherapy agent[7]. When combined with EndoTAG-1, the dual approach targets both the tumor cells (via standard paclitaxel) and the tumor's blood vessels (via EndoTAG-1). The combination has shown promising activity in cancer treatment, particularly for triple-negative breast cancer (TNBC). Clinical trials have evaluated this combination therapy, showing improvements in progression-free survival compared to monotherapies in metastatic TNBC and high pathological complete response rates in neoadjuvant HER2-negative breast cancer, particularly in TNBC patients. The combination has shown a manageable safety profile, although grade 3 hypersensitivity reactions to EndoTAG-1 and other adverse events like neutropenia have been reported.
