Enfortumab vedotin + carboplatin is a combination regimen of two agents: **enfortumab vedotin**, an antibody-drug conjugate that targets Nectin-4 with a cytotoxic payload of monomethyl auristatin E (MMAE), and **carboplatin**, a platinum-based chemotherapeutic. Enfortumab vedotin delivers MMAE to Nectin-4–expressing cancer cells, leading to microtubule disruption, cell cycle arrest, and apoptosis. Carboplatin causes DNA crosslinking and apoptosis by forming platinum-DNA adducts. This combination is primarily under clinical investigation for the treatment of advanced or metastatic urothelial carcinoma in patients unable to receive cisplatin or immunotherapy.
