**Enfortumab vedotin + gemcitabine** is an investigational combination therapy consisting of two distinct drugs: enfortumab vedotin, an antibody-drug conjugate (ADC) targeting Nectin-4 with a microtubule inhibitor payload (monomethyl auristatin E), and gemcitabine, a nucleoside analog antimetabolite chemotherapeutic. Enfortumab vedotin delivers its cytotoxic payload selectively to Nectin-4-expressing tumor cells, causing cell cycle arrest and apoptosis, while gemcitabine interferes with DNA synthesis, further inhibiting tumor cell proliferation. This combination is being studied primarily for advanced or metastatic urothelial carcinoma and has been used as a comparator regimen in trials evaluating newer combinations for this indication[1][3][6][4].
