Overview

Enobosarm + pembrolizumab is a combination therapy evaluated primarily for the treatment of androgen receptor–positive (AR+) metastatic triple-negative breast cancer (mTNBC). Enobosarm is a nonsteroidal selective androgen receptor modulator (SARM) that targets the androgen receptor, while pembrolizumab is a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD‑1). The rationale for this combination stems from preclinical evidence suggesting that AR-targeted therapy may enhance antitumor immunity when combined with immune checkpoint blockade. In phase II clinical trials, this regimen demonstrated good tolerability and modest clinical benefit in heavily pretreated patients with AR+ mTNBC[1][2][3][4][5][7][8].
