Overview

ENPP1 inhibitors (Leuven CD3) are a series of small molecule compounds developed by the Leuven Centre for Drug Design and Discovery (CD3) in collaboration with KU Leuven researchers. These inhibitors target Ectonucleotide Pyrophosphatase/Phosphodiesterase 1 (ENPP1), an enzyme that plays a critical role in immune evasion by degrading cyclic GMP-AMP (cGAMP). cGAMP is a key secondary messenger that activates the Stimulator of Interferon Genes (STING) pathway, which is essential for initiating an innate immune response against tumors. By blocking ENPP1, these small molecules prevent the hydrolysis of cGAMP, thereby enhancing STING activation, increasing the production of Type I interferons, and promoting the recruitment and activation of T-cells within the tumor microenvironment. This approach aims to convert immunologically "cold" tumors into "hot" tumors, potentially improving the efficacy of existing checkpoint inhibitors. The program is currently in the preclinical stage of development for applications in immune oncology and various cancers.
