A combination regimen composed of **ensituximab**, a chimeric IgG1 monoclonal antibody that targets a variant of the tumor-associated antigen MUC5AC with antibody-dependent cellular cytotoxicity; **gemcitabine**, a nucleoside analog that inhibits DNA synthesis; and **nab-paclitaxel**, an albumin-bound form of the microtubule inhibitor paclitaxel. Ensituximab is under clinical investigation for solid tumors expressing MUC5AC, particularly refractory metastatic colorectal and pancreatic cancers. Gemcitabine and nab-paclitaxel together are a standard chemotherapy backbone for pancreatic cancer. This investigational combination aims to augment antitumor immune responses with ensituximab and cytotoxicity via gemcitabine and nab-paclitaxel.
