Overview

Entecavir + tenofovir is a fixed-dose combination of two oral antiviral agents used primarily for the treatment of chronic hepatitis B virus (HBV) infection, especially in patients with previous nucleoside/nucleotide analogue treatment failure or multidrug-resistant HBV. Entecavir is a guanosine nucleoside analogue that inhibits HBV DNA polymerase, thereby blocking viral replication. Tenofovir is an adenosine nucleotide analogue that also inhibits viral reverse transcriptase and DNA polymerase, causing chain termination and inhibition of viral synthesis. The combination leverages their high genetic barriers to resistance and non-overlapping resistance profiles to provide effective suppression of HBV replication in patients with complex resistance mutations. This regimen has demonstrated efficacy and safety as rescue therapy for chronic hepatitis B, particularly in those with prior antiviral drug resistance[1][3][4][5].
