This is a combination regimen of three antiviral agents used primarily for the treatment of chronic hepatitis B virus (HBV) infection. Entecavir is a guanosine nucleoside analogue that selectively inhibits HBV polymerase, blocking all three steps in the viral replication process[6][7]. Tenofovir, typically as tenofovir disoproxil fumarate or tenofovir alafenamide, is a nucleotide reverse transcriptase inhibitor that also targets HBV polymerase and suppresses viral DNA synthesis[3]. Peginterferon alfa-2b is a recombinant form of interferon with polyethylene glycol conjugation; it acts by binding to type 1 interferon receptors and activating the JAK/STAT pathway, leading to an enhanced innate antiviral response[5]. The combination leverages different mechanisms—direct inhibition of viral replication by nucleoside/nucleotide analogues and immune modulation by peginterferon—to improve rates of hepatitis B surface antigen (HBsAg) clearance compared to monotherapy. This approach may be used in various strategies: simultaneous initiation ("de novo"), sequential ("switch-to"), or add-on therapy for patients already on nucleos(t)ide analogues[1][2][8].
