This combination therapy consists of high-dose interleukin-2 (aldesleukin) and entinostat, a selective class I histone deacetylase (HDAC) inhibitor. Interleukin-2 is a cytokine that promotes the proliferation and activation of T cells and natural killer cells, while entinostat is an epigenetic modulator designed to enhance the immune response by reducing the number and suppressive function of regulatory T cells (Tregs) and myeloid-derived suppressor cells (MDSCs). The regimen is primarily being investigated by Dr. Roberto Pili for the treatment of advanced or metastatic clear cell renal cell carcinoma, particularly in patients who have progressed on prior immune checkpoint inhibitors. The combination aims to overcome resistance to immunotherapy by modulating the tumor microenvironment.
