Entospletinib + idelalisib is an oral combination therapy comprised of two investigational small molecule inhibitors: entospletinib, a selective spleen tyrosine kinase (SYK) inhibitor, and idelalisib, a selective phosphatidylinositol 3-kinase delta (PI3Kδ) inhibitor. This combination was studied in relapsed or refractory chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma (NHL), with the rationale that dual inhibition of B-cell receptor pathway targets would provide synergistic antitumor activity. However, a phase 2 study was terminated early due to high rates of severe pneumonitis, including fatalities, despite some clinical responses[1][4].
