Drug intelligence / Profile preview
Envafolimab + albumin-bound paclitaxel + platinum is an investigational chemoimmunotherapy regimen combining the subcutaneous PD-L1 inhibitor envafolimab with the microtubule inhibitor albumin-bound paclitaxel (nab-paclitaxel) and a platinum-based chemotherapeutic agent (such as cisplatin or carboplatin). Envafolimab is a single-domain antibody that blocks the interaction between PD-L1 and PD-1, thereby restoring T-cell-mediated anti-tumor immune responses. Albumin-bound paclitaxel promotes tubulin polymerization and inhibits mitosis, while the platinum agent binds to DNA to form crosslinks, interfering with replication and transcription. This combination is primarily being evaluated in clinical trials as a neoadjuvant or conversion therapy for patients with locally advanced esophageal squamous cell carcinoma (ESCC) and head and neck squamous cell carcinoma (HNSCC), aiming to achieve synergistic anti-tumor efficacy and improve pathological complete response (pCR) rates.
