Envafolimab + axitinib is a combination therapy being investigated for the treatment of advanced clear cell renal cell carcinoma (ccRCC). Envafolimab is a recombinant humanized single-domain antibody (nanobody) that acts as a programmed death-ligand 1 (PD-L1) inhibitor; it is unique as the first PD-L1 inhibitor formulated for subcutaneous injection. Axitinib is an oral, potent, and selective tyrosine kinase inhibitor of vascular endothelial growth factor receptors (VEGFR-1, VEGFR-2, and VEGFR-3). By combining immune checkpoint blockade with anti-angiogenic therapy, this regimen aims to enhance anti-tumor efficacy in treatment-naive patients with metastatic or unresectable kidney cancer. The combination is currently being evaluated in a Phase II clinical study led by Peking Union Medical College Hospital.
