Overview

Envafolimab + ipilimumab is a **combination of two monoclonal antibodies** for cancer immunotherapy, currently investigational and studied in advanced or metastatic undifferentiated pleomorphic sarcoma (UPS) and myxofibrosarcoma (MFS) that have progressed on prior chemotherapy[1][3][7]. Envafolimab is a single-domain anti-PD-L1 antibody administered subcutaneously; it blocks the interaction of programmed death-ligand 1 (PD-L1) with its receptor PD-1 on T cells, preventing tumor immune evasion and stimulating immune attack against cancer[4]. Ipilimumab is a human IgG1 monoclonal antibody that binds the checkpoint molecule cytotoxic T-lymphocyte antigen-4 (CTLA-4), preventing inhibition of T-cell responses and thereby activating T cells to attack tumors[6]. The two drugs together aim to synergistically enhance T-cell mediated anti-tumor responses by overriding multiple immune checkpoints.
