Overview

Envafolimab + lenvatinib + paclitaxel-albumin is an investigational combination regimen comprising three agents with distinct mechanisms of action, studied primarily in advanced solid tumors such as gastric/gastroesophageal junction adenocarcinoma. Envafolimab is a subcutaneously administered humanized single-domain anti–PD-L1 monoclonal antibody that blocks the interaction between PD-L1 and its receptor, restoring T-cell mediated antitumor immunity. Lenvatinib is an oral small molecule tyrosine kinase inhibitor targeting multiple receptors including VEGFR, FGFR, PDGFRα, RET, and KIT to inhibit tumor angiogenesis and proliferation. Paclitaxel-albumin (nab-paclitaxel) is a nanoparticle formulation of the microtubule-stabilizing chemotherapeutic agent paclitaxel bound to human serum albumin for improved solubility and delivery. This combination aims to synergistically enhance antitumor efficacy by integrating immune checkpoint inhibition, antiangiogenic therapy, and cytotoxic chemotherapy[1][2][7].
