Envafolimab + trastuzumab + docetaxel is an investigational combination therapy being evaluated for the treatment of HER2-positive breast cancer. The regimen consists of **envafolimab** (KN035), a unique subcutaneous PD-L1 single-domain antibody Fc fusion protein; **trastuzumab**, a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2); and **docetaxel**, a taxane-based chemotherapy agent. Envafolimab is designed to block the interaction between PD-L1 and PD-1, thereby restoring T-cell-mediated anti-tumor immune responses. Trastuzumab inhibits tumor cell proliferation by binding to the extracellular domain of HER2 and mediating antibody-dependent cellular cytotoxicity (ADCC). Docetaxel acts by stabilizing microtubules and inhibiting their disassembly, leading to cell cycle arrest and apoptosis. This combination is currently being studied in Phase II clinical trials sponsored by Sun Yat-Sen Memorial Hospital to assess its efficacy and safety in patients with HER2-positive disease.
