Overview

Envolimab + lenvatinib is a combination therapy consisting of envolimab, a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), and lenvatinib, an oral multi-tyrosine kinase inhibitor. Envolimab acts as an immune checkpoint inhibitor by blocking PD-1, thereby enhancing T-cell mediated antitumor immune responses. Lenvatinib inhibits multiple receptor tyrosine kinases involved in tumor angiogenesis and proliferation, including vascular endothelial growth factor receptors (VEGFR) 1–3, fibroblast growth factor receptors (FGFR) 1–4, platelet-derived growth factor receptor alpha (PDGFRα), RET, and KIT. The combination is being investigated primarily for the treatment of advanced or unresectable hepatocellular carcinoma (HCC). This dual approach aims to both suppress tumor-driven angiogenesis/proliferation and enhance the host immune response against cancer cells.
