enzalutamide + CC-115 is a combination therapy investigated for the treatment of metastatic castration-resistant prostate cancer (mCRPC). Enzalutamide is a second-generation androgen receptor (AR) signaling inhibitor that blocks androgen binding, nuclear translocation, and DNA binding. CC-115 is an orally bioavailable, dual inhibitor of the mechanistic target of rapamycin (mTOR) complexes 1 and 2 and the DNA-dependent protein kinase (DNA-PK). The combination aims to address resistance mechanisms in prostate cancer, where the PI3K/AKT/mTOR pathway is frequently activated (often via PTEN loss) and compensates for AR inhibition. By simultaneously targeting AR signaling, mTOR-mediated survival pathways, and DNA-PK-mediated DNA repair, the combination seeks to enhance anti-tumor activity and delay the progression of castration-resistant disease.
