Overview

enzalutamide + PSA-TRICOM is a combination therapy being evaluated for the treatment of non-metastatic castration-sensitive prostate cancer (nmCSPC). The regimen consists of enzalutamide (Xtandi), a potent androgen receptor inhibitor, and PSA-TRICOM (PROSTVAC), a therapeutic cancer vaccine. Enzalutamide works by competitively inhibiting androgen binding to the androgen receptor, as well as inhibiting its nuclear translocation and DNA binding, thereby suppressing the growth of prostate cancer cells. PSA-TRICOM is a viral vector-based immunotherapy that utilizes a heterologous prime-boost strategy with recombinant vaccinia (PROSTVAC-V) and fowlpox (PROSTVAC-F) viruses. Both vectors express human prostate-specific antigen (PSA) and a triad of costimulatory molecules (B7.1, ICAM-1, and LFA-3, collectively known as TRICOM) designed to enhance the activation and recruitment of T-cells against PSA-expressing tumor cells. This combination is primarily being studied in trials sponsored by the National Cancer Institute (NCI).
