**EO-3021 + dostarlimab** is an investigational combination therapy being evaluated for advanced solid tumors, specifically in cancers expressing Claudin 18.2 such as gastric and gastroesophageal junction (GEJ) cancer. EO-3021 is an antibody-drug conjugate (ADC) that targets Claudin 18.2, delivering the cytotoxic payload monomethyl auristatin E (MMAE) via a cleavable linker. It retains antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), leading to direct tumor cell killing as well as a bystander effect in the tumor microenvironment. Dostarlimab (brand name Jemperli) is a monoclonal antibody targeting the programmed death-1 (PD-1) receptor; it blocks the PD-1 pathway, thereby enhancing T cell-mediated anti-tumor immunity. The combination is designed to target cancer cells with EO-3021, while also releasing immune inhibition with dostarlimab, synergistically promoting anti-tumor activity. The combination is currently in early-phase clinical development.
