EO2040 + nivolumab is an investigational combination therapy for colorectal cancer, specifically targeting patients with circulating tumor DNA (ctDNA)-defined minimal residual disease (MRD) after completion of standard curative therapy. EO2040 is a synthetic, off-the-shelf peptide-based immunotherapy developed by Enterome. It consists of two OncoMimics peptides—microbial-derived peptides that mimic CD8 HLA-A2 epitopes from the tumor-associated antigens FOXM1 and BIRC5—and includes universal cancer peptide 2 (UCP2), a helper peptide representing a CD4+ epitope. The combination with nivolumab, an immune checkpoint inhibitor targeting PD-1, aims to enhance anti-tumor immune responses by both stimulating T-cell activity against specific tumor antigens and blocking inhibitory signaling in T cells. This regimen is being evaluated in the Phase 2 CLAUDE trial for safety, tolerability, immunogenicity, and preliminary efficacy as measured by ctDNA clearance and absence of radiographic recurrence[1][2][3][4][5].
