Drug intelligence / Profile preview
EO4010 + nivolumab is an investigational combination immunotherapy regimen being evaluated for the treatment of microsatellite stable (MSS) and mismatch repair proficient (pMMR) metastatic colorectal cancer (mCRC). EO4010 is an off-the-shelf therapeutic cancer vaccine developed by Enterome, consisting of five synthetic microbial-derived peptides (EO2317, EO2318, OMP10, OMP11, and OMP12) that mimic HLA-A2 restricted CD8+ T-cell epitopes of five tumor-associated antigens (TAAs) upregulated in colorectal cancer: BIRC5 (survivin), FOXM1, UBE2C, CDC20, and KIF2C. It also includes the helper peptide Universal Cancer Peptide 2 (UCP2) to stimulate CD4+ T cells. Nivolumab (Opdivo) is a programmed death-1 (PD-1) blocking monoclonal antibody developed by Bristol Myers Squibb. By combining EO4010's ability to expand tumor-specific CD8+ T cells with nivolumab's checkpoint inhibition, the regimen aims to overcome immune tolerance and enhance anti-tumor activity in immunologically cold MSS colorectal tumors.
